I am working with featured images. they’re linked with posts that contain vimeo-embed links, like that:

<iframe src=”//player.vimeo.com/video/77196393?byline=0&color=ffffff” width=”500″ height=”211″ frameborder=”0″ webkitallowfullscreen mozallowfullscreen allowfullscreen></iframe>

is it possible to open these links with the lightbox instead of opening a new page?
or am i totally wrong with that?

Yes. If you embedd your video not in an iframe way but using a link or shortcode (as described in the codex http://codex.wordpress.org/Embeds) it should automatically work the way you’d like to.
